DESCRIPTION:(Re) Defining Myeloid Cell States and Function in Alzheimer’s Disease \nJoin the Center for the Neurobiology of Learning and Memory in a virtual colloquium with Dr. Beth Stevens\, Associate Professor of Neurology at Harvard University. \nAbstract: Emerging evidence from genetic association studies shows that dysregulation of the brain’s immune system plays a key role in the onset and progression of neurodegenerative disease. Our research aims to identify and validate disease mechanisms informed by human genetics and identify functional and mechanistic markers of myeloid and glial cells in detrimental states\, before cognitive decline\, that can be readily detected through imaging and fluid biomarkers. Using single-cell RNA sequencing\, spatial transcriptomics and proteomic approaches we identified molecular signatures that can be used to localize and monitor distinct microglia functional states in human and mouse brain. Our results show that microglia assume diverse functional states in development\, aging and injury\, including populations corresponding to known microglial functions including proliferation\, migration\, inflammation\, and synaptic phagocytosis. In addition\, we identified several innate immune pathways by which microglia recognize and prune synapses during development and in models of disease\, including the classical complement cascade. Illuminating the mechanisms by which developing synaptic circuits are sculpted is providing important insight on understanding how to protect synapses in Alzheimer’s and other neurodegenerative diseases of synaptic dysfunction.

DESCRIPTION:C. elegans Processes Sensory Information to Choose Between Freeloading and Self-defense Strategies \nJoin the UCI Department of Developmental and Cell Biology in a virtual seminar with Dr. Javier Apfield\, Assistant Professor of the College of Science of Northeastern University. \nAbstract:\nHydrogen peroxide is the preeminent chemical weapon that organisms use for combat. Individual cells rely on conserved defenses to prevent and repair peroxide-induced damage\, but whether similar defenses might be coordinated across cells in animals remains poorly understood. In this seminar\, I will discuss our recent findings showing that in the nematode Caenorhabditis elegans a neuronal circuit processes information perceived by two sensory neurons to control the induction of hydrogen-peroxide defenses in target tissues. We found that catalases produced by Escherichia coli\, the nematode’s food source\, can deplete hydrogen peroxide from the local environment and thereby protect the nematodes. In the presence of E. coli\, the nematode’s neurons signal via TGFβ-insulin/IGF1 relay to target tissues that they need not induce catalases and other hydrogen-peroxide defenses. DESCRIPTION:Dopamine waves as a mechanism for spatiotemporal credit assignment. \nJoin the UC Irvine Center for Addiction Neuroscience in a virtual ICAN Seminar featuring Arif Hamid\, PhD\, from Brown University. \nAbstract\nSignificant evidence supports the view that dopamine shapes reward-learning by encoding prediction errors. However\, it is unknown whether dopamine decision-signals are tailored to the functional specialization of target regions. In this talk\, I will report on a novel set of wave-like spatiotemporal activation patterns in dopamine axons (and release) across the dorsal striatum that switch between different activational motifs. At reward delivery\, waves are altered by task demands\, propagating from dorsomedial to dorsolateral striatum during instrumental contingencies\, and in the opponent direction when rewards are independent of animal behavior. I will demonstrate that our results are consistent with a computational architecture in which striatal dopamine signals are sculpted by inference about agency\, and provide evidence for a spatiotemporally “vectorized” role of dopamine in credit assignment directed toward specialized “expert” striatal subregions.

DESCRIPTION:Development and diversity of neural cell types in the septum \nThe CNLM will be hosting Dr. Corey Harwell\, from the Department of Neurobiology of Harvard Medical School\, in a virtual colloquium where he will present his latest research. \nAbstract: The septum is ventral forebrain structure responsible for the regulation of emotional states including anxiety\, fear and depression. The septum contains an extremely diverse array of short- and long-range projecting GABAergic neurons distributed across its two histological subdivisions\, the medial and lateral septal nuclei. Both of these regions are heavily interconnected with other brain areas. It is currently unclear how septal neuronal diversity and circuit wiring are specified during development. Using molecular genetics and single-cell/nuclei RNA-seq we have begun to unravel the developmental logic for producing diverse neural cell types in the septum. Our future work is focused on understanding the specific contribution of developmentally specified neural cell types to the regulation of internal states carried out by the septum.

DESCRIPTION:Parallel Dopamine Circuits Mediate Unique Aspects of Cocaine-Induced Behavioral Plasticity  \n  \nPlease join the UCI School of Pharmacy & Pharmaceutical Sciences in a virtual seminar featuring Kevin Beier\, Ph.D. \n  \nAbstract: \nMy research is focused on identifying how experience modulates activity dynamics in neural circuits\, both acutely and chronically. I aim to develop new methods and use these in combination with modern techniques such as optogenetics and in-vivo calcium imaging to understand the sources of adaptive and maladaptive plasticity that drive normal and pathophysiological behaviors. These include reward/aversion processing in normal appetitive behaviors\, drug addiction\, neurodegeneration\, and neurodevelopmental disorders such as autism. I will also employ cutting-edge viral-genetic methods to map brain state-dependent activity in connected neural ensembles\, and use this information to infer how our brain state drives us to perform particular behaviors. I also aim to take novel approaches to interrogating the sources of individual behavioral variation in neuropsychiatric conditions such as addiction\, anxiety\, and depression.

DESCRIPTION:Department of Pharmaceutical Sciences Research Seminar Series:  \nIntergenerational Trauma: Two-hit Mechanism and Early Preventive Interventions \nPlease join the Department of Pharmaceutical Sciences in a research seminar featuring UC Irvine Associate Professor\, Dr. Amal Alachkar. \nAbstract: \nThe deleterious impacts of existential trauma during pregnancy are doubled as they affect the mental health of two generations (the mothers’ and the unborn offspring). In my lab we try to establish early biomarkers for identifying individuals at risk\, and for predicting novel treatment targets at the earliest practicable ages. Our studies show that trauma exposure in mice during pregnancy induces in the offspring social deficits and depressive-like behavior through divergent and convergent mechanisms of both in utero and early-life parenting environments. Molecular mechanisms of intergenerational trauma transmission involve stress- and hypoxia-response energy pathways\, especially mitochondrial ATP production\, and epigenetic processes. We establish 2-Hydroxyglutaric acid as an early predictive biomarker for trauma-induced behavioral deficits. Early but not late pharmacological interventions with acetyl-L-carnitine supplementation\, produces long-lasting protection against the behavioral deficits associated with intergenerational trauma.
